I just went to Sakura Castle and I saw that there was a ~lvl30 Toad Boss. I was wondering if the real one still exists.

Yup. Just go to Mushroom shrine and talk to those two bums lazing around. They'll take you to the original Ninja Castle. You'll still need to get through the jq in order to get to him.
