Yes as long as you don't hit 500/500 points, or else it doesn't work. Example would be killing Zakum after gathering 400/500 points. The amount from Zakum will give 275 ambition points. So it will then be 675/500.

Pro-tip:

Get as close as you can to 500, then kill a hard boss like czak or PB that gives a lot of ambition; it'll break through.
